Spherical human basophil is rendered as a circulating granulocyte, with a centrally placed, bilobed to S-shaped nucleus partially obscured by dense cytoplasmic granules. Coarse basophilic secretory granules occupy most of the peripheral cytoplasm and crowd the perinuclear region, creating the classic appearance in which nuclear contours are harder to trace than in a neutrophil. A thin rim of plasma membrane defines the cell boundary, with granule-rich cytoplasm lying immediately deep to it. Basophils matter because their granules store histamine, heparin, and preformed mediators that drive immediate hypersensitivity reactions after IgE cross-linking at the cell surface, a mechanism shared conceptually with mast cells but encountered in the peripheral blood film. In routine hematology, confusing a basophil with a neutrophil is a common learner error, and this view emphasizes the discriminators: fewer nuclear lobes, darker coarse granules, and the tendency of granules to overlie the nucleus. Clinically, basophilia is a classic peripheral smear clue in myeloproliferative neoplasms such as chronic myeloid leukemia, and accurate visual pattern recognition supports that teaching point.
