Description
Rendered as a three-dimensional human mast cell, the model centers on an irregular plasma membrane with multiple slender filopodia projecting outward into the extracellular space. A large, ovoid nucleus occupies a central to slightly eccentric position within the cytoplasm, while numerous membrane-bound secretory granules cluster in the perinuclear cytoplasm and extend toward the peripheral cytoplasm. Granules lie deep to the cell cortex, ready for trafficking to the membrane, and their dense packing gives the cell its classic granulocyte-like appearance despite mast cells being tissue-resident immune cells rather than circulating granulocytes. Mast cells sit at the junction of immunology and clinical allergy because their granules contain preformed mediators such as histamine, heparin, and proteases (for example tryptase), which can be discharged by IgE-dependent activation through Fc epsilon RI. Degranulation at the cell surface drives the immediate phase of type I hypersensitivity, from urticaria and allergic rhinitis to bronchospasm and systemic anaphylaxis, and the perinuclear granule reservoir helps explain the rapid kinetics of mediator release.
